Hello, may I ask this - NMR experiments/pulse sequences are available to assign aromatic sidechains - Phe, Tyr, Trp? We'd like to try 2D and 3D.

What's available in the vendor-specific libraries?

There are standard hcch cosy and tocsy experiments: hcchcogp3d, hcchdigp3d, and hcchdigp3d2. And there are the "music" pulse sequences: music_fhyw_3d is for Phe/His/Tyr or Trp. I don't know if there are any other specialized ones.

Lewis Kay's lab published (HB)CB(CGCD)HD and (HB)CB(CGCDCE)HE experiments quite some time ago. Neither is all that sensitive, but they're only 2D experiments. You can also look for strong intra-residue NOEs between the ring protons and HA/HB, although that requires more caution.

Once you've correlated HD and/or HE to the backbone, you can get the rest of the ring protons with COSY, TOCSY, and NOESY, then the carbons with HSQC/HMQC.

I've done these and they complemented a 13C NOESY quite well. I was able to get a good 80% of my aromatics signals seen by these experiments - roseyp (Feb 22 '11 at 09:38)

There a number of the C13 detect experiments that might help, the simplest might be the 13C-13C COSY experiment in the aromatic region, see http://onlinelibrary.wiley.com/doi/10.1002/cmr.a.20109/pdf

Of course you have to have access to a carbon detect cryoprobe
