Hi, someone asked a similar question before, but I'd like to know:

Are there readily available varian pulse sequences that help assign aromatic side-chains? Maybe there is something like that in Biopack or elsewhere?

Hi,

In VJ/BioPack you can go to the Experiments Menu/Protein Triple Resonance Experiments/Aromatics you'll find the entries for the two experiments mentioned by Evgeny. These will run the setup macros so you can run just like any other BioPack Experiment.

Hi there,

I've done a quick look at the /vnmr/psglib directory and found these:

  • hbcbcgcdceheA.c (correlates shifts CB with HD and HE)
  • hbcbcgcdhdA.c (correlates CB with HD)

Both are 2D experiments and seem to be using Biopack parameters.

Once you assign HD, HCCH-TOCSY should help you assign the remaining resonances of the aromatic side-chains (but you might have to optimize parameters of HCCH-TOCSY for the aromatics).
